DESCRIPTION:
Differential gene expression was evaluated between male Sprague Dawley rats in the Oprm1 knockdown (a mu opioid receptor) and control groups. Effects of Oprm1 downregulation in the medial preoptic area (MPOA) on patterns of gene expression in the MPOA were examined using RNA-Seq. Paired-end 2x150 bp sequencing was performed, using standard SBS chemistry (v3) on an Illumina NovaSeq6000 sequencer. We conducted differential gene expression analysis of RNA-Seq data using the EdgeR Bioconductor Package, v. 3.9. The final samples for RNA-Seq analysis were Scr shRNA control (n=7); Oprm1 shRNA knockdown (n=5). In the behavior experiments, Oprm1 downregulation produced less social play behavior in male rats.
