Gene expression in 212,713 ventral midbrain single nuclei from 95 individuals with history of opioid misuse, and individuals without drug exposure. Chronic exposure to opioids was not associated with change in proportions of glial and neuronal subtypes, however glial transcriptomes were broadly altered, involving 9.5 - 6.2% of expressed genes within microglia, oligodendrocytes, and astrocytes. Genes associated with activation of the immune response including interferon, NFkB signaling, and cell motility pathways were upregulated, contrasting with down-regulated expression of synaptic signaling and plasticity genes in ventral midbrain non-dopaminergic neurons. Ventral midbrain transcriptomic reprogramming in the context of chronic opioid exposure included 325 genes that previous genome-wide studies had linked to risk of substance use traits in the broader population.

To understand mechanisms and identify potential targets for intervention in the current crisis of opioid use disorder (OUD), postmortem brains represent an under-utilized resource. By comparing transcript profiles of normal mucosa and CRC samples in two independent sample sets, covering a total of 20 normal mucosa and 424 adenocarcinoma samples, we identified 51 TFs that showed a significant (P<10−4, Benjamini and Hochberg corrected) fold change (of at least 2) from normal to cancer in both data sets.
The production of 12 out of 27 measured factors was induced by CEsHUT including IL-1β, TNF and IL-1Ra. In contrast to sIL-1Ra production, that of IL-1β and TNF was inhibited by HDL, corroborating previous results. In addition, CEsHUT induced monocytes to produce factors involved in their localization, survival and differentiation such as CCL5 (RANTES), CCL2 (MCP-1), interferon-γ (IFNγ), granulocyte-macrophage colony-stimulating factor (GM-CSF), and macrophage-CSF (M-CSF). The production of the latter was moderate and it was not affected by HDL.
